She was playing to the audience in the room. Unfortunately for her, 15 million households were watching it live, not to count the folks who have watched it since. Lots of candidates, good and bad, say things that are stupid at the time. Most don't think it will leave certain rooms, and others may not think that the mic is on. Regardless, I don't think that one comment makes or breaks her as a candidate in the primaries or the general. I can think of other reasons to question her viability as a candidate - questioning her judgement based on the email issue, for example. I don't think that one matters. A lot of folks realize that this is the time in the campaigns when candidates play to their base. They may not hold it against her in a general.
