Not necessarily.
There will be several factors that will be in play, mainly:
1) What the economy looks like in '24. People vote with their wallets. If inflation, shortages, and supply-chain issues remain a problem or get worse, Trump would have an easier route back in - just repeat Clinton's mantra "It's the economy, stupid" with "Are you better off than you were four years ago". This will resonate with a lot of Americans who will hold their nose and vote for Trump.
2) Biden's age - assuming he's the Democratic nominee. If he's not, the Dems would have to nominate someone who could beat Trump head on - and that person would have to be a moderate like Biden. The only problem is who would fit that bill?
3) Whether Biden keep Harris as VP. Biden's poll numbers are pretty weak. Harris' are in the tank. She's a huge albatross around his neck. The thought of her being President would actually be worse than Trump to many people - even moderate Dems. However, if he cans her it would cause a backlash in the Dem fringe that could cost him black support if its played out right by the Republicans.
4) Biden continuously putting out idiotic suggestions for bills that the majority of Americans don't support.
5) Biden's inability to fix problems here at home. From the Colorado being run dry to inner city crime, to immigration problems and drug deaths and unemployment, all these things are going to be hung around his neck.
Plenty more, but I think to cast Trump as a defeated candidate already is a bit premature. Way too much time left to go until '24 to do that.
