The nearest star is 4 light years away. The fastest we've ever gone in space (unmanned) is 0.02% the speed of light. So right there it would take 5000 years to get there with that speed. But let's say we could get to to 10% of light speed... that's still 40 years.
The scary thing I think Hank is referring to is that we could - and perhaps are - getting so technically advanced that if your society can figure out how to build a space ship that goes that fast, your society also probably figured out how to quickly and easily destroy yourself. For example, with advanced AI, could pretty much anybody figure out how to destroy life on this planet?
