Yes, I will be the first to admit that it didn't pan out with respect to the continuation of DNL343 for the treatment of patients with Amyotrophic Lateral Sclerosis [ALS]. The phase 2/3 HEALEY ALS Platform Trial Regimen G was trying to see if there were at least some type of biomarker findings [neurofilament light chain] that would allow it to continue this specific program. However, even if you set this aside, it has made headway with the FDA agreeing to a Priority Review filing for potential Accelerated Approval of tividenofusp alfa for the treatment of patients with MPS II.
Where the risk still remains is whether or not it is actually going to be allowed Accelerated Approval based on the surrogate biomarker endpoint of reduction of CSF HS. If this doesn't happen, then it is going to have to rely on its confirmatory study, which is likely to take a longer time to complete. The beauty of this biotech is being able to expand the use of its ETV platform to target a host of many other lysosomal storage disorders. I went over some of these above, like FTD-GRN, Pompe, Gaucher, and MPS I.
With the expected Priority Review PDUFA date of April 5, 2026, of tividenofusp alfa for the treatment of patients with MPS II, plus the ability to expand with expected data at the 2026 WORLDSymposium from the phase 1/2 study using DNL126 to treat patients with MPS IIIA, I believe that DNLI stock should remain with a Strong Buy rating.

Bought RKT and FIGR last year at their lows and doubled my money already. I think both are gonna keep running as long as there are no rate surprises. FIGR is one of the most interesting stocks in my portfolio b/c they are one of the only real companies that’s commercializing blockchain. Not like these other BS “projects” you hear about. It’s legit.

Actually, the data suggests investing a lump sum even at all-time highs is the best choice. I believe this works best ~60% of the time. Over 98% of the time a new all-time high leads to another all-time high. This makes logical sense, but obviously the emotional aspect of this is more difficult.
My advice, you can make more than one choice. For example, half in the market now, have keep in cash until you feel comfortable. Something like that?
I believe there is a time/movement component to this. A new ATH hits another new ATH prior to it dipping X% or something like that.
